华佗自幼聪慧过人、醉心医学，因缘际遇下，拜名医张仲景为师。一次，关云长带同乡求医，佗发现此人的病症有异，不是普通伤寒，故将《伤寒论》和《滋阴论》结合，成功研究出治疗瘟疫之法。惜百姓迷信鬼神之说，加上小人得志，对佗诸多加害，令其惹上杀身之祸。仲景之长女张妙心，与佗两情相悦，决心下嫁并随夫逃亡。

佗移居许昌后有医无类，不论病得多重的人也全力拯救，结果往往失败，被评为“医死人大夫”，生活每况愈下。后佗替曹操的幼儿医好多年顽疾，得峰赏识，封为太医院监，著手撰写医学大全。好景不常，操的奸雄本色渐露，命佗制毒药杀害关之士兵，佗不耻操所为，投靠关。操头风病发作，恼羞成怒，以斩杀太医院内三百余人作胁，迫佗返回许昌为其治病，佗深知此行凶多吉少，莫非这就是命运的安排？
