Haru Tezuka's parents run a bar in Kamata, Tokyo. She is honest and bright, but she can't lie which sometimes causes her to get in trouble. Haru finds it difficult to get a job. Haru then notices an application guideline from airline company ANA (All Nippon Airways). Haru, who never thought of working in the airline industry, decides to take a test for the airline company. She first attends a company presentation by ANA. There, she sees elite persons who have dreamed of becoming pilots for lengthy periods of time and airplane enthusiasts. She becomes overwhelmed by them. Haru is indecisive about whether she wants to go through with it, but she manages to barely pass her test. Now, Haru wants to become a pilot, but what awaits for her is harsh training.

没什么远大理想，天真烂漫、直率不会撒谎的性格让刚大学毕业的手冢晴（堀北真希饰）在找工作时屡屡被拒。就在这时她看到了ANA的招聘简章，并接受了从未想过会涉足的航空界的测试。一起参加公司说明会的，都是长年梦想当飞行员的精英和飞机爱好者。小晴在考试中勉强及格，开始走上了女飞行员的道路，然而等待着她的却是超乎寻常的严格的训练。

主角手塚晴（堀北真希飾）是東京蒲田居酒屋店主的女兒，因為性格直率不擅巧言令色，導致求職接連碰壁。一次偶然的機會中參加航空公司ANA的機師考試，意外獲得錄取，在教官的嚴格指導下，一步步的朝向機師的目標邁進。

Les parents de Haru Tezuka gèrent un izakaya à Kamata. Haru est brillante, honnête et spontanée, cette dernière caractéristique lui causant des difficultés pour trouver un emploi. Après plusieurs échecs elle remarque une brochure de candidature pour devenir pilote au sein de la compagnie aérienne ANA. Haru, qui n'a jamais pris l'avion, décide de tenter sa chance et découvre sa vocation mais sa formation sera rude.

『ミス・パイロット』で、堀北真希が演じるのは、女性パイロットという狭き門に挑む主人公・手塚晴（てづか・はる）。晴は、東京・蒲田にある居酒屋の娘。天真爛漫、素直でウソがつけない性格が災いし、手当り次第に受けた会社は全部不採用。就職活動に苦戦し、万策尽きた晴の目に入ったのは、ANAの募集要項だった。晴は、全く考えもしなかった航空業界の試験を受けてみることに。足を運んだ会社説明会では長年パイロットを夢見てきたエリートや飛行機マニアに圧倒されながらも、あれよあれよという間にギリギリ合格し、女性パイロットへの道を歩むことになる。しかし、そこに待っていたのは、予想を超えた厳しい訓練の数々だった。苦難に立ち向かいながら、大きな夢に向かって切磋琢磨する晴の姿を中心に、彼女が副操縦士になるための4年の歳月を追う。
