The Ten Commandments volgt het verhaal uit het Bijbelboek Exodus over de geboorte van Mozes, zijn jeugd aan het hof van de farao, zijn leiderschap in de verlossing uit de slavernij en uittocht uit Egypte van het Joodse volk, de ontvangst van de Tien Geboden en zijn opvolging door Jozua.
Bruce Nolan is een nieuwsverslaggever met ambitie, maar ziet de rest van de wereld zich hogerop werken terwijl hijzelf achterblijft in het middelmatige. Na wederom een dag vol tegenslagen verwijt Bruce God nalatigheid, omdat Hij nooit naar hem luistert. God hoort dit blijkbaar wel en geeft Bruce Zijn krachten onder het motto: kun jij het beter? Deze taak blijkt echter best lastig te zijn.
Mozes werd als kind te vondeling gelegd door zijn wanhopige moeder en werd gevonden door de dochter van de Farao. Hij groeide op bij de Egyptische koninklijke familie samen met toekomstige Farao Ramses II.

De Egyptische farao Seti geeft het bevel om alle Hebreeuwse baby's te verdrinken. Een wanhopige moeder plaatst haar zoon in een mandje, die terecht komt bij de koningin. Moses wordt opgevoed als een broer van de troonopvolger Ramses. Jaren later worden de broers van elkaar gescheiden wanneer Moses achter zijn ware achtergrond komt.
Een gewone man wordt door God geroepen om het onmogelijke te doen in deze liefdevol vertelde productie van het oudtestamentische verhaal. Wanneer de farao van Egypte de strop om zijn joodse slaven begint te spannen, leidt Mozes (Ben Kingsley) hen naar de vrijheid.
Het eerste deel van deze film vertelt het verhaal van Mozes die de Joden van Egypte naar het Beloofde Land, zijn ontvangst van de gedenkplaten en de aanbidding van het gouden kalf. Het tweede deel laat de uitwerking van de geboden in het normale leven in San Francisco zien.
In een aantal sketches wordt de draak gestoken met verschillende periodes uit de geschiedenis van de mensheid, zoals de tijd van de holbewoners, de Romeinse tijd en de Franse Revolutie.
