Da NASA-direktør og tidligere astronaut Jo Fowler (Halle Berry) erfarer, at månen inden for kort tid vil blive slået ud af sin bane, og således føre til udslettelse af jorden og menneskeheden, tager hun en ukonventionel beslutning: Jo Fowley søger hjælp hos såvel en tidligere astronaut og en konspirationsteoretiker. Trods faren for eget liv begiver disse usandsynlige helte sig snart ud på en umulig sidste mission i rummet, kun for at opdage, at vores måne slet ikke er, hvad vi tror, den er...

Videnskabsmanden Robert Neville er helt alene i storbyen efter at en dødbringende virus har udslettet resten af New Yorks befolkning. Han prøver desperat at komme i kontakt med andre overlevende ved hjælp af sin radio, men der er tilsyneladende ikke andre. I hvert fald ikke om dagen. Men om natten kommer de inficerede mutanter frem. Og de holder øje med Neville.

Et sælsomt syn møder drengen Pazu, da den jævnaldrende Sheeta langsomt daler ned fra himlen med en magisk amulet om halsen. Det viser sig, at både Pazu og Sheeta drømmer om at finde Laputa, det svævende slot. Den forældreløse Sheeta håber at finde sandheden om sin familie på Laputa, men regeringens agenter og en gruppe pirater er også på jagt efter det sagnomspundne slot.

På vegne af det britiske imperium drager en af verdens sidste store opdagelsesrejsende, britiske Percy Fawcett, i starten af det 20. århundrede til Amazonas jungle for at kortlægge området mellem Brasilien, Peru og Bolivia. I junglen finder han spor på en hidtil ukendt, højt udviklet civilisation. På trods af, at de etablerede videnskabsmænd latterliggør ham og hans tese om et teknisk avanceret urfolk i junglen, bliver udforskningen til en besættelse for ham. Han tager nu tilbage til regnskoven for at bevise, at han har ret.

Et team af videnskabsmænd, der arbejder på at rejse en sunket russisk atomubåd på en havplatform ud for Miamis kyst, Florida, afgraver et gammelt atlantisk levn fra havbunden og henter en ekspert til at finde ud af det. In San Francisco, well-known sportsman Kay Hoog announces to a club that he has found a message in a bottle with a map drawn by a Harvard professor who has gone missing. The map tells of a lost Incan civilization that possesses an immense treasure. Hoog immediately plans an expedition to find it. But Lio Sha, the head of a criminal organization known as the Spiders, is determined to get the treasure for herself and plans a rival expedition.

The film presents compelling evidence that Altlantis wasn't so far away after all, but what it omits is just as compelling. Although Plato’s account of Atlantian masonry, consisting of red, white and black stones, was a visually perfect match for the modern walls of the Minoan excavation site favored by the film, and his account of a sea made impassable by small islands of mud could, in fact, be a description of the rafts of pumice left by the catastrophic eruption of an ancient volcano there, little mention is made of Plato's specific account of where Atlantis was or the common root that links Atlantis to the Atlantic ocean. Also omitted is Plato's chronology placing the sinking of Atlantis in the same time frame as the end of the last ice age which caused the inundation of huge expanses of once fertile lands.
