Yu-Gi-Oh! Duel Monsters, noto fuori dal Giappone semplicemente con il nome Yu-Gi-Oh!, è il secondo adattamento anime della serie. È stato prodotto da Nihon Ad Systems e TV Tokyo, mentre l'animazione per lo spettacolo è stata realizzata da Studio Gallop.

La storia inizia a partire dall'ottavo capitolo del manga. Rispetto al manga originale la serie è stata liberamente adattata. La serie presenta diverse differenze rispetto al manga e alla serie prodotta da Toei e si concentra principalmente sul gioco di carte Duel Monsters, in sintonia con la vita reale di Yu-Gi-Oh! Gioco di carte collezionabili. La serie è andata in onda in Giappone su TV Tokyo tra il 18 aprile 2000 e il 29 settembre 2004, per 236 episodi.

Ginga Hagane (Gingka nell'edizione italiana) assieme a Pegasus, il suo beyblade, decide di viaggiare intorno al mondo per vendicare suo padre e riprendere L-Drago: il bey proibito rubato da Doji e Ryuga. Durante il suo viaggio finisce per fermarsi in una città in cui si farà degli amici (Kenta, Madoka, ecc.) e dei rivali.
