首集《死亡筆記本》中，天才大學生－夜神月（藤原龍也飾），撿到死神掉落在人間的筆記本，原先想利用死亡筆記本殺盡壞人，到後來走火入魔，演變成L與夜神月間的懸疑鬥智對決。緊接著，第二本筆記本從天而降…。 夜神月以「協助逮捕奇樂」之名，加入了搜查本部，Ｌ則虎視眈眈地等待著。此時，與路克想法截然不同的另一名死神雷姆，在地上掉落了另一本死亡筆記本。而持有這本筆記本的，正是崇拜奇樂的偶像女星彌海砂（戶田惠理香飾）。變成第二個奇樂的彌海砂得到「死神之眼」的力量，這是拿來對付Ｌ的最強武器。幸好Ｌ制敵機先，將海砂以奇樂嫌疑犯的身份監禁在奇樂對策室裡。 除了舊班底藤原龍也、松山健一等的主要角色外，本片還加入了戶田惠理香、片瀨那奈等女星。重新解明的死亡筆記本使用規則，讓天才們的算計失控，能看透一切的將會是誰？
