徳川家康が天下を統一した後の1614年。忍者の二大勢力である伊賀と甲賀の両勢力は、初代服部半蔵のおきてにより長らく戦うことを禁じられていた。そんな中、伊賀当主の孫娘・朧と甲賀当主の後継ぎである弦之介は、互いの素性を知らぬまま出会い、恋に落ちる。一方、伊賀と甲賀の両勢力は、家康の命によりそれぞれの精鋭5人ずつを戦わせ、どちらが勝ち残るかで次期将軍を決めるという忍術合戦を強いられることになる。
