Arkeologiparin ohella miehistön ytimeen kuuluvat yhtiön etua raivokkaasti valvova Meredith Vickers sekä Weylandin itsensä suunnittelema superälykäs, ihmistä täysin muistuttava David-androidi, jonka inhimillisyys ei kuitenkaan kata kaikkia tunnetiloja. Prometheus vie tutkimusryhmän universumin pimeimpään ja syrjäisimpään kolkkaan, jossa heitä odottaa taistelu koko ihmiskunnan tulevaisuuden puolesta.
