Faber College heeft een studentenvereniging die iedereen lid maakt die maar zin heeft om lid te worden. Bij een andere studentenvereniging zitten alleen rijke leden en dus word je niet zomaar lid van deze vereniging. Natuurlijk botst het tussen deze twee verenigingen.
De film speelt zich af tussen studenten in hun eerste jaar op college. Rachel is de leidster van de Kappa Phi sociëteit en de nr. 1 op de 'hottie' ranglijst van de universiteitswebsite. Haar status wordt echter bedreigd door nieuwelinge Sydney White, een vriendelijke brunette die bovendien bij Rachels vriendje Blaine in de smaak valt.
Van Wilder (Bennett) is onderweg naar de universiteit en is van plan daar ook alles op z'n kop te zetten, op de "Van Wilder"-manier. Maar wat hij niet weet is dat hij één van de conservatiefste universiteiten heeft uitgekozen.
