Drive My Car berkisah tentang seorang pria bernama Yusuke Kafuku (Hidetoshi Nishijima) kehilangan istrinya secara mendadak. Dua tahun setelah kematian sang istri, ia memutuskan untuk melanjutkan karirnya sebagai aktor dan sutradara panggung teater. Pada suatu kesempatan, ia menerima tawaran untuk sebagai pengarah produksi Uncle Vanya di sebuah festival teater di Hiroshima. Sebagai bentuk jaminan keselamatan, seorang pengemudi wanita bernama Misaki Watari (Toko Miura) ditugaskan untuk mengantar Yusuke ke Hiroshima. Perjalanan Yusuke ke Hiroshima dimulai dengan si pendiam Misaki dan Saab 900 merahnya. Saat pemutaran perdana produksi mendekat, situasi tidak mengenakan terjadi.
