Seit Jahren schon tobt der Krieg gegen die außerirdischen Gamilon und fordert hohe Verluste. Die Menschheit verliert den Krieg gegen die Gamilas und steht kurz vor der endgültigen Auslöschung. Die Yamato hat Iscandar und die große magellansche Wolke verlassen und befindet sich auf dem ersehnten Rückweg zur Erde, als sie von einem aggressiven neuen Feind angegriffen wird: Den Gatlantern! Diese konnten sich gerade erst von der Gewaltherrschaft der Gamilas befreien, streben nun aber selbst die galaktische Hoheit an. Da die Yamato nicht mehr über ihre volle Stärke verfügt, muss die Crew auf eine List zurückgreifen. Doch an dem Ort, an dem sie sich wiederfinden, scheinen die Gesetze der Physik und auch die der Logik nicht zu gelten…
Als bösartige Außerirdische, bekannt als die Gamilon, auf der Erde nukleare Verwüstung anrichten, rüstet eine Gruppe von Überlebenden das japanische Schlachtschiff Yamato für intergalaktische Reisen um und begibt sich auf eine Mission, um einen Neutralisator zu finden, der die Strahlung aus der Erdatmosphäre tilgen soll.
The Galman Empire is destroyed but the planet Galmania is not, by a chance collision of galaxies. The Bolar Federation worlds including Planet Bolar are destroyed. The Yamato, back under the command of Captain Okita, encounters the planet Denguil too late to save its humanoid civilization from being flooded by the water planet Aquarius. The surviving Denguil, a warrior race who believe only the strong should survive, plan to use Aquarius to flood Earth and destroy humanity, in order to create a new home for their race.

While Dessler gathers together the shattered remnants of his Gamilus Empire, new forces are at work in the universe that could augur in another conflict. Seems the Black Star Empire has invaded Dessler's homeworld and is decimating it, all the while gathering power for its war machine.
The anime's story is set in the year 2202, three years after the Yamato's return from the planet Iscandar. The people of Earth restored their planet with the Cosmo Reverse System, and they signed a peace treaty with Garmillas. In addition to reconstruction, Earth developed a new defense fleet that includes the state-of-the-art battleship Andromeda. The Earth goes down a path of military expansion, despite Starsha Iscandar's wishes. The Yamato helps accomplish this so-called peace, but it comes at the price of many invaluable sacrifices. The goddess Teresa, who prays for tranquility in space, calls the Yamato to a new voyage. The threat of Gatlantis is sweeping over the universe and approaching Earth.

Space Battleship Yamato is a Japanese science fiction anime series featuring an eponymous spacecraft. It is also known to English-speaking audiences as Space Cruiser Yamato; an English-dubbed and heavily edited version of the series was broadcast on North American and Australian television as Star Blazers. The first two seasons of this version were broadcast in Greece in 1981-82 as Διαστημόπλοιο Αργώ. An Italian-language version was also broadcast under the name Star Blazers in Italy, and a Portuguese-language version was successfully shown in Brazil under the title Patrulha Estelar and Viaje a la Ultima Galaxia or Astronave Intrepido in Spain and Latin America.
It is a seminal series in the history of anime, marking a turn towards more complex serious works and influencing works such as Mobile Suit Gundam and Neon Genesis Evangelion; Hideaki Anno has ranked Yamato his favorite anime and credited it with sparking his interest in anime.
